Abstract

Endurance exercise enhances 5-hydroxytryptamine synthesis and tryptophan hydroxylase expression in dorsal raphe of 6-hydroxydopamine-induced Parkinson's rats. Exercise Science, 16(2): 177-185, 2007. In addition to the loss of dopamine neurons in Parkinson's disease, there are many evidences on the dysfunction of 5-hydroxytryptamine (serotonin) system and degeneration of 5-hydroxytryptamine neurons in the dorsal raphe. In the present study, we investigated the effect of treadmill exercise on the synthesis of 5-hydroxytryptamine and on the expression of tryptophan hydroxylase in the dorsal raphe by using immunohistochemistry. The lesion of nigrostriatal dopamine pathway in rats was made by unilaterally injection of 6-hydroxydopamine into the striatum. The rats in the exercise group were made to run on the treadmill for 30 minutes once a day during 14 consecutive days after 6-hydroxydopamine injection. Two weeks after injection of 6-hydroxydopamine, 5- hydroxytryptamine synthesis and tryptophan hydroxylase expression in the dorsal raphe were decreased. It indicates that 6-hydroxydopamine- induced destruction of dopamine system in the striatum impaired serotonergic neural system in the dorsal raphe. However, treadmill running increased 5-hydroxytryptamine synthesis and tryptophan hydroxylase expression in the dorsal raphe of 6-hydroxydopamine- injection rats. The present study revealed that endurance exercise is effective for the symptom improvement in Parkinson's disease patients.* This work was supported by the Korea Research Foundation Grant funded by the Korea Government (MOEHRD)(KRF-2005-041-G00120).
