Abstract

The purpose of this study was to investigate the effect of exercise training and quercetin supplementation with low fat diet on pro-inflammation and Th1/Th2 cytokine production by antigen-stimulation after one bout maximal exercise on immunocompetence in high fat diet mice. To accomplish the purpose of this study, C57BL/6 male mice were fed high fat diet (45% fat diet) for 8 weeks. Then these mice were divided into 3 groups: HLC (high fat diet and low fat diet for 8 weeks as the control, n=8), HLQ (high fat diet and low fat diet for 8 weeks with quercetin supplementation, n=8) or HLE (high fat diet and low fat diet for 8 weeks with moderate exercise training, n=8). Quercetin (10 mg/kg) was administrated once a day, 5 days/week for 8 weeks. Exercise training was performed for 8 weeks on a treadmill running for 30-60 min/day at 10-22 m/min, 0% grade, 5 days/week. Just before killing the mice, all the mice performed maximal exercise 60 minutes at a speed of 25 m/min (VO2max 80-85%) as an exercise-induced fatigue. Peritoneal macrophage was collected and stimulated by lipopolysacharide (0, 0.5, 1.0 µg/ml) for 24 h and splenocyte stimulated by concanavalin A (0, 5, 10 µg/ml) for 48 h. Then pro-inflammatory cytokine (TNF-α, IL-6, IL-1β) and Th1(IL-2, IFN-γ)/Th2(IL-4, IL-10) cytokine were measured by ELISA kit. As a result, body weight, TNF-α and IL-6 were significantly reduced in HLE compared to HLC and HLQ. Also, IL-10 was increased in HLE compared to HLC and HLQ. These findings suggest that moderate exercise training has beneficial effects on body weight and immunocompetence in high fat diet mice.
