초록
As cell-based research and development continues to grow, a variety of cell culture technologies are emerging. In particular, as the scope of research in the field of biomimetic science has expanded recently, the development of 3D cell culture methods has become another research focus. In this study, we developed a culture support ware that can be effectively applied to scaffold-based cell culture.First, we used a 3D printer to fabricate a scaffold using polycarbolactone (PCL), and designed a mold for a 3D cell culture insert suitable for scaffold mounting. The mold design consisted of two types: connected and disconnected structure, and the molding and processing conditions were reviewed through simulation. The insert well was molded using polystyrene material, which is non-cytotoxic and widely used in cell culture vessels and medical devices. We then mounted the scaffold into the insertion well and cultured adipose-derived mesenchymal stem cells(ADMSCs) three-dimensionally to verify the effectiveness of scaffold-based cell culture. As a result, ADMSCs grew better when the scaffold was placed in the insert well than when the scaffold was placed directly on the culture dish, and cell surface markers were expressed at levels similar to those in the basic culture method. From a usability perspective, the connected type is more convenient than the detachable type. However, the mold design allows for the formation of both types, enhancing the efficiency of the mold. Therefore, it is expected that applying verification through preliminary simulation, and injection molding technology to the development of cell culture ware will facilitate the development of cell culture methods.
