초록
An effective mass production system via somatic embryogenesis was successfully established for a new thornless cultivar of Aralia elata (Miq.) Seem. ‘Seochun’. The stem and leaf explants were treated with various concentrations (0, 0.5, 1.0, 2.0, 3.0 and 4.0 mg/L) of 2,4- dichlorophenoxyacetic acid (2,4-D) to induce somatic embryos. The results demonstrated that indirect somatic embryos were only effectively induced in 9 stem explants (30%) treated with 0.5 mg/L 2,4-D, which were part of the 12 stem explants (40%) that also showed callus formation under the same treatment. In contrast, all leaf explants across all 2,4-D concentrations produced only callus without any somatic embryo formation. Additional experiments were conducted using different concentrations of gibberellic acid (GA3) to enhance in vitro growth performance of plants derived from somatic embryos. The findings indicated that treatment with 1.0 mg/L GA3 significantly improved growth, resulting in an average stem length of 4.6 ± 1.0 cm and a root length of 8.0 ± 2.8 cm, outperforming both untreated plants and those treated with 0.5 mg/L GA3. Herein, a comprehensive production system was optimized, including somatic embryogenesis induction, in vitro propagation, and successful acclimatization for cultivation. The results provide a novel foundation for the stable mass production and propagation of thornless cultivar of Aralia elata ‘Seochun’ offering significant potential for commercial applications and forestry advancement.
