Abstract
In this paper, we confirmed the anionic surfactant dioctyl sodium sulfosuccinate(DOSS), with natural yellow 3(NY3) targeting protein in blood, can fix bloody fingermarks on non-porous surfaces. Also, we suggested an optimal concentration of DOSS for NY3 to minimize blood smudging. Crime scene investigators are using blood fixatives like 2% 5-sulfosalicylic acid (SSA) to fix protein in blood before applying protein-targeting dyes to enhance bloody fingermarks. We pretreated bloody fingermarks as various solutions followed by amido black(AB) and acid yellow 7(AY7) application. Results showed that the 2% DOSS solution, along with ethanol and acetic acid, could reduce smudging of bloody fingermarks. Enhancing 1:8 diluted blood with 0~5% range of DOSS containing NY3 resulted in increased fluorescence with higher DOSS concentrations, but smudging was observed at 0~3% DOSS concentration. Enhancing bloody fingermarks on OHP films and slide glasses with NY3 showed smudging at 0~3% and 0~2% DOSS concentrations, respectively. Comparative evaluation between the previously suggested 2% DOSS concentration and the 4% DOSS concentration from this experiment revealed that the 4% DOSS concentration yielded better results in minimizing smudging, especially for larger blood volumes or blood contaminated surfaces. Therefore, to minimize blood smudging, it is recommended to use NY3 with a DOSS concentration of 4% or higher.
