Abstract

Mosaic loss of Y chromosome (mLOY) is a form of somatic mosaicism, characterized by the coexistence of genetically distinct cell populations within a single individual, resulting from the loss of the Y chromosome in a subset of male somatic cells. mLOY can occur across all age groups and typically presents as a 45,X/46,XY karyotype. In this study, we report a case of mLOY observed in blood, skin cell swabs (dorsum of the hands, neck), and buccal swab samples collected from an elderly male decedent. The proportion of Y DNA was determined through quantitative analysis of specific regions of the Y chromosome and by comparing the peak heights of amelogenin and DYS391 loci using a commercial STR kit. Decreased Y DNA levels were observed in all samples, with blood and skin cell samples showing significantly lower Y DNA proportions compared to buccal samples. These findings suggest the potential for errors in interpreting contributor ratios and profiles during the analysis of mixed DNA profiles in forensic casework, highlighting the need for caution.
