Abstract
Gender identification is the most basic process in STR analysis, but there are several risk factors that can cause serious confusion in forensic gender identification. In the previous paper, a part of the Y-STR was found despite being female DNA, and a very rare interchromosomal insertional translocation of the Y-chromosome-P4-fragment (529 kb) was reported. In this paper, the sequence was analyzed by Linked-read sequencing (10x Genomics) analysis in female autopsy DNA showing the same phenomenon. The reason why the same Y-STR DNA type (10,18 in DYS385, 11 in DYS460) was detected in both cases was interpreted as being inherited from common ancestor rather than recurrent translocations caused by de novo. Three possible causes were suggested for the unexpected result of partial detection of Y-STR despite female DNA. First, human error, second, medical intervention, and third, genetic abnormality. When Y-STR is detected in DNA evidence that is not of male origin, criteria for interpreting it are presented.
