Abstract

Examination of 16 bloodstain samples collected from a 71-year-old male homicide victim revealed diverse Amelogenin gene locus expression patterns despite originating from the same individual, including female-type, mixed-type, and normal male-type patterns. DNA quantification analysis confirmed highly variable male : female DNA ratios across samples, ranging from 1 : 2,981.6 to 1 : 1.3, indicating non-uniform Y chromosome loss. STR analysis using both GlobalFiler kit and PowerPlex Fusion kit consistently demonstrated these patterns, with the signal intensity of Y-chromosome markers DYS391 and Y-Indel also showing significant variation between samples. Identical Y-STR profiles detected across all samples are estimated that the discrepancies in Amelogenin results stemmed from mLOY (mosaic loss of Y chromosome) rather than contamination or mixed DNA. This mLOY phenomenon is presumed to occur through mechanisms involving incomplete separation of the Y chromosome during cell division or selective loss during cell death processes. Unlike previous studies focusing on primer binding site mutations or gene deletions, our research showed an acquired, heterogeneous Y chromosome loss phenomenon frequently occurring in elderly males.
