Abstract
Rye genome- and chromosome-specific DNA markers were selected to easily identify the existence of rye chromatin inthe wheat genome by RAPD analysis. Among 260 decamer primers used, five primers, namely, OPC01, OPF07, OPF11, OPH09,and OPH16 amplified rye genome- and chromosome-specific DNA fragments. The OPC01 marker was amplified from the tem-plate DNA of rye chromosome 7, while OPF07 marker could be used for the selection of lines having rye chromosome 1. BothOPF11 and OPH09 markers were amplified from rye chromosome 6. The OPH16 marker was obtained from rye chromosome 1,4, 6 and 7 respectively. The sizes of rye chromosome-specific markers of OPC01, OPF07, OPF11, OPH09 and OPH16 were 1207bp, 1987 bp, 1225 bp, 1354 bp and 1772 bp, respectively. Five rye chromosome-specific markers identified in this study can beused effectively for the detection of rye chromatin as well as the assignment of introduced rye chromosome in breeding programthrough triticalewheat hybridization.
