Abstract

Chrornium{VJ) is wideJy used in industry and m에 or metals used in the nickeJ-chrornium a1Joys of most or바lOdonitic appJiances, partial denture but chrornium{VJ) is a potent inducer of tumors in human. The mechanism of cytotoxicity and carcinogenesis associated with these compounds did not understand well. The aims of this study was canied out to detect the mechanism 01 cytotoxicity and carcinogenesis of chrornium{VJ) in cultured NIH3T3 fibrobJasts. The cytotoxicity of chromium{VJ), gallic acid and peroxidase was evaluated by the MTT and XTT assays. A1so, antitoxic effects of galJic acid, peroxidase and gallic acid pJus peroxidase against IG-JÜ of chrornium{VJ) were evaJuated by the same methods. The results were as follows ; l. The cytotoxicity of chrornium trioxide (Cr03) and sodium dicromate dihydrate (Na~Cr2Ü7' 2H20) were highly toxic by Borenfreumd et al’. ICso of chrornium trioxide and sodium dichromate dihydrate by the MTT assay were 18.4 )1M and 18.0 )1M, respectively and the values of ICso by the XTT merhod were deterrnined at 33.9 )1M and 42.5 )1M, respectively. 2. The cytotoxicity of gallic acid was nontoxic. However peroxidase was moderately toxic IG-:Il of gallic acid and peroxidase by MTT and XTr assay were 2,552.4 )1M, 2,여5‘6 )1M, 718.7 )1M and 622.7 )1M, respectively. 3. The antitoxic effect of galJic acid against ICso of chromium trioxide and sodium dichromate dihydrate were statisticalJy significant increased in 50 )1M, but antitoxic effects of 100 )1M was more deceased than that of 50 )1M. 4. The antitoxic effec!s of peroxidase against IG-JÜ of chrornium trioxide and sodium dichromate dihydrate were statistically significant by a dose dependent manner. 5. The antitoxic effects of galJic acid pJus peroxidase against IG-JÜ of chrornium trioxide and sodium dichromate dihydrate was more incrased than that of galJic acid or peroxidase treated only. These resuJts suggest that gallic acid or peroxidase may be decrease the cytotoxicity of chrornium{VI) and gallic acid pJus peroxidase are more decrease than that of galJic acid or peroxidase treated onJy.
