Abstract
Hexavalent chromium[chromium{Vl)] is commonly used in industrial chrome painting, welding, metal finishes, steel manufacturing, alloy, cast iron and wood treatment, and is a proven toxin, mutagen and carcinogen. The mechanistic cytotoxicity of chromium{Vl) is not completely understood, but, a large number of studies demonstrated that chromium{Vll induces oxidative stress, DNA danmge, apoptotic cell death and altered gene expression. This study was carried out to detect the compounds which could decrease the cytotoxicity of chromium{Vl) in cultured NH3T3 fibrobasts. The Cytotoxicitv of sodium dichromate dihvdrate, chromium tIioxide and ponciIin itself was evaluated bv MTT assay for cell viability and XTT assay for cell adhesion activity, Also, antitoxic effects of poncirin against cytotoxicity of s여ium dichromate dihydrate and chromium trioxide was evaluated by same methods. The results were as follows : 1. The 10>J of sodium dichromate dihydrate by MTT and XTT assay were 44.0 11M and 69.5 11M, respectively. 2‘ ηJe 10>J of chromium trioxide by MTT and XTT assay were 51.2 11M and 69.2 11M, respectively, 3, The 10>J of poncirin itself by MTT and XTT assay were 2,915,7 11M and 2,725,3 11M, respectively. 4, Poncirin was statistically significant decreased cytotoxicity of sodium dichromate dihydrate from 1 11M to 50 11M concentration by dose dependent manner, 5, Poncirin was statistically significant decreased cytotoxicity of chromium trioxide from 1 uM to 25 uM concentration by dose dependent manner. These results suggest that poncirin may decrease cytotoxicity of sodium dichromate dihydrate and chromium trioxide,
