초록

The apical region of rodent incisors has been proposed as a stem cell niche that enables continuous growth. In this study, to identify transcriptional genes involved in dental hard tissue cell differentiation, we compared the expression profiles of ninety genes between undifferentiated cells in the apical region and differentiated cells in the supra-apical region of rat incisors using a PCR array. In our model, mRNAs encoding CAP, enamelin, ameloblastin, amelogenin, and Dspp were exclusively expressed in the supra-apical region. Vdr, Rb1, and Tcf7L2 were upregulated by more than twofold in the supra-apical region. In contrast, Tcfe2a, Ldha, Ezh2, Klk10, Pitx2, Fos, and Sp1 were upregulated by twofold or greater in the apical region. Foxa2, Gata6, Hoxc6, Olig2, and Wt1 were detected only in the supra-apical region, while five genes including Hoxa1 and Zic2 were exclusively found in the apical region. These findings identify candidate transcriptional genes associated with the differentiation of dental hard tissue cells, including cementoblasts. Further studies are needed to characterize their specific roles in tooth development, including cementogenesis.
