Abstract

Silver impregnation techniques are frequently used for demonstration of argyrophilic or argentaffin substances in histopathology. The conventional methods for silver staining generally require long incubation of lhr to 24hr or more at 60 t. The purpose of this study'was carried out to investigate the effect of microwave irradiation for rapid silver impregnation. Silver impregnation techniques used in this study were Masson - Fontana, periodic acid methenamine- silver(PAM), Grocott’s methenamine-silver(GMS), and AgNORs staining. Tissue samples fixed by immersion in 10 % neutral buffered formalin and processed for routine light microscopy. Tissue sections immersed in silver solution were irradiated to 30 seconds interval using microwave oven, and then incubated for 5 minutes. We could be shortened the impregnation time within lmin incubation after 3min(65 ℃), 5min(80 ℃), and 4min(75℃)-irradiation by microwave in Fontana - Masson, PAM, and GMS staining, repectively. These staining activity were also nearly equal to conventional method. However, AgNORs staining was somewhat inferior in staining activity. In addition, tissue structures were well-preserved in all samples. This result suggested that microwave irradiation was useful to shorten impregnation time in some silver staining.
