Abstract
The Neo-Timm’s su1phide-silver method has frequently been used for histochemical 10 cahzation of heavy metals including zinc in various tissues. However, this method has the shortcoming of being difficult for general application , because should be perfused through the thoracic artery with fixatives saturated with H2S gas. We were attempted to evaluate the possibility for general application using the modification of the NeoTimm’s method. Tissues resected were fixed in 100% ethanol saturated with H2S gas at room temperature for 48 hr, and then processed according to routine procedure for light microscopy. Strongly positive staining was found in the horny layer of epidermis , connective tissue of dermis and in hair roots of skin, medullary portion of kidney, zona fasciculata of adrenal, and tunica albunigea and several cellular components of the spermatogenic chain of testis. Chelation with diethyldithiocarbamate(DEDTC) prevented this sta.ining. This result suggested that modified Neo-Timm's sulphide-silver method used in this study is a simple, rapid, and reliable staining for the localization of heavy metal, mainly zinc, in these organs.
