면역세포화학적 반응의 전자현미경적 관찰을 위한 전포매법과 후포매법의 비교

Abstract

Immuno-electron microscopy is a standard method for subcellular localization of specific antigens in tissue or cell preparation. We compared pre-embedding and post-embedding methods to detect cytokeratin and proliferating cell nuclearantigen(PCNA) using immunocytochemistry at electron microscopic level the results were as follows; 1. By pre-embedding method, positive reaction for cytokeratin was observed in the cytoplasm, but not for PCNA. 2. By post-embedding method using Unicryl resin, gold particles of cytokeratin and in both cytoplasm and nucleus of cells PCNA were stained by usual immunohistochemistry. In conclusion, the best results were obtained by post - embedding method using Uncryl resin. We suggest the post-embedding method is useful both in light and electron microscopy.
