Abstract
This study evaluated the in vitro biological activities of Diospyros lotus extract, specifically its antioxidant, anti-inflammatory, skin barrier-enhancing, and wound healing properties. The extract significantly protected HaCaT keratinocytes from oxidative stress induced by H2O2, confirming its strong antioxidant and cytoprotective effects. High-performance liquid chromatography (HPLC) analysis identified gallic acid (2.291 mg/g) as the major phenolic compound in the seed extract, followed by rutin, quercetin, luteolin, and astragalin. Cytotoxicity assays conducted on RAW 264.7 macrophages and HaCaT cells demonstrated that concentrations up to 400 μg/mL were non-toxic. In LPS-stimulated macrophages, the extract significantly reduced the production of nitric oxide (NO) and interleukin-6 (IL-6) in a dose-dependent manner, indicating robust anti-inflammatory activity. Additionally, treatment of HaCaT cells increased the mRNA expression of epidermal barrier-related genes, including filaggrin, involucrin, and loricrin. In a wound healing assay, D. lotus extracts, particularly the leaf fraction (DLL), notably accelerated keratinocyte migration and wound closure. These findings suggest that D. lotus extract has multifunctional biological activities, highlighting its potential as a natural cosmetic ingredient for skin protection, inflammation control, and barrier recovery.
