초록

Phytochromes, a family of light-sensing proteins, are widespread in a broad rangeof organisms, including plants, algae, cyanobacteria, and even non-photosynthetic bacteria. Deinococcus radiodurans, which is well known for its extraordinary resistance to γ-radiation,contains bacterial phytochrome photoreceptor (BphP). D. radiodurans BphP (DrBphP) is knownto dephosphorylate its cognate response regulator DrBphR in response to light and to contributeto mitomycin C (MMC) resistance. In this study, we investigated transcriptional changes in D. radiodurans bphP bphR double mutant strain (ΔbphPR) to identify genes affected by the DrBphP/Rsystem. Microarray analysis showed that the expression of dr_0802, which encodes a putative antisigmafactor, and dr_1168, which encodes a Nudix hydrolase, increased in ΔbphPR. Since a geneencoding the alternative sigma factor Sig2 immediately precedes dr_0802, the predicted DR_0802product was named Asf2 (anti-sigma factor 2) here. We constructed sig2 and asf2 mutant strainsand examined their survival using plates supplemented with MMC under light conditions. A lackof either Sig2 or Asf2 did not cause a change in resistance to MMC compared to the wild type (WT),i.e., there was no difference in survival between WT and mutants. These results suggest that Sig2and Asf2 are not involved in the DrBphP/R signaling pathway related to MMC resistance.
