Abstract

The study investigates whether the composition of amino acids in ruminant microbial, differs according to different energy source feeds used for Korean beef cattle (Hanwoo) finishing diets. This experiment analyzed ruminal fermentation patterns, microbial protein synthesis (MPS), microbial amino acid composition, and the microbial community after 72 h continuous culture in vitro rumen fermentation usingcorn grain (T1), rice bran (T2), wheat grain (T3), and wheat bran (T4). The pH in the culture was maintained at 5.5~7.0 during theincubation in all treatment. The total VFA concentration was highest (29.93 mM) in T4 and lowest (23.13 mM) in T1 (p<0.05). TheA (acetate) :P (propionate) ratio was highest (2.81) in T4 and lowest (1.48) T1 (p<0.05). The butyrate was lowest (2.37 mM) T1 andhighest (4.37~4.58 mM) in T2, T3, T4 (p<0.05). The MPS was highest (332.5 mg/L) T2 and (320.2 mg/L) T3, lowest (137.5 mg/L)T1 and (154.2 mg/L) T4 (p<0.05). The rumen microbial community analysis by DGGE showed a 57.5% similarity or less between treatments. The total amino acid concentrations in rumen microorganisms was highest (31.59 %) T1 and (31.33 %) T3, lowest (20.09 %) T2 (p<0.05). This was difference to the total amino acid concentration in the feed. Therefore, this study confirmed the different energy feeds for Hanwooaffected rumen fermentation patterns, MPS changing the microbial community and amino acid content of rumen microorganisms.
