Abstract
Transient oxidative stress induced by exercise and chronic oxidative stress observed in diabetic patients are known to cause cellular damage, inflammatory responses, and mitochondrial dysfunction, thereby impeding muscle recovery and metabolic regulation during rehabilitation. Such oxidative damage, accompanied by an imbalance in the antioxidant defense system, exacerbates glucose dysregulation; therefore, the discovery of effective antioxidant and antidiabetic agents is crucial for nutritional management in rehabilitation. In this study, the antioxidant and antidiabetic activities of a mixed extract (HCML) composed of barley (Hordeum vulgare) grains, chicory (Cichorium intybus) roots, bitter melon (Momordica charantia) fruit, and banaba (Lagerstroemia speciosa) leaves were evaluated. The antioxidant effects of HCML were assessed using DPPH and ABTS radical scavenging assays, as well as a reducing power analysis. The results showed a concentration-dependent increase in antioxidant activity, and 500 μg/mL of HCML exhibited a DPPH radical scavenging effect comparable to that of ascorbic acid. Furthermore, α-amylase and α-glucosidase inhibitory assays demonstrated that HCML inhibited digestive enzymes in a concentration-dependent manner. Notably, 500 μg/mL HCML showed an α-glucosidase inhibitory effect similar to that of acarbose, suggesting its potential to suppress postprandial blood glucose elevation. These findings indicate that HCML is a multifunctional bioactive material capable of simultaneously alleviating oxidative stress and improving carbohydrate metabolism. In particular, it may contribute to maintaining metabolic homeostasis through oxidative stress reduction and glycemic control during post-exercise recovery or in individuals with chronic metabolic disorders. Therefore, this study provides fundamental data supporting the development of functional beverages or nutritional supplements applicable to rehabilitation and clinical nutrition management.
