Diabetes mellitus is a metabolic disorder characterized by impaired regulation of blood glucose levels, and its prevalence has been rapidly increasing worldwide. Although various antidiabetic drugs have been developed, many food-derived substances have long been known to exert hypoglycemic effects. Among these, freeze-dried powder of 3rd-day, 5th-instar silkworm larvae (FDSP) and Banaba leaf extract (BLE) have been approved by the Korean Ministry of Food and Drug Safety as individually recognized and notified health functional foods for blood glucose control, respectively. In this study, we developed a novel manufacturing process to enhance the release of the active compounds 1-deoxynojirimycin (1-DNJ) from FDSP and corosolic acid from BLE. The 1-DNJ extract from FDSP (1DNJE) produced through this process exhibited superior hypoglycemic activity compared with conventional FDSP. BLE also demonstrated a notable blood-glucose-lowering effect. Interestingly, the combined treatment of the 1DNJE and BLE showed a hypoglycemic effect comparable to that of metformin, a standard antidiabetic drug. The combined effect is likely due to their distinct mechanisms: 1-DNJ inhibits α-glucosidase, thereby preventing the hydrolysis of polysaccharides into monosaccharides, whereas corosolic acid promotes glucose uptake in muscle and adipose tissues and enhances hepatic glucose utilization. Therefore, the combination of 1DNJE and BLE produces an additive effect on glucose reduction, suggesting its potential as a natural alternative or supplement for diabetes management.
