초록
An efficient regeneration protocol for stem discculture of Populus euramericana, which is important speciesfor bioenergy resource in agroforestry, was established. Thenumber of explants that were obtained and the number ofexplants that regenerated varied with the genotypes. However,in all the genotypes, stem disc culture produced more regeneratedshoots than did in axillary bud culture. A comparisonof the effects of cytokinin type and concentration on shootregeneration in different explants (i.e., petiole, leaf, and rootsegments of P. euramericana) revealed that a concentrationof 0.002 mg l-1 thidiazuron (TDZ) used on petiole segmentsresulted in the greatest shoot regeneration (95.83%). Thehormonal requirements for the greatest shoot regeneration inthe three explant types varied. Different concentrations ofAgNO3 and CoCl2 were added separately to the medium tostop the yellowing and subsequent necrosis of the regeneratedshoots. Lower concentrations (3 and 5 mg l-1) of these compoundsimproved shoot regeneration and elongation, compared withthe control. The in vitro-regenerated shoots were transferredto rooting medium and subsequently acclimatized. The highlyefficient regeneration system of P. euramericana reportedhere can be used for mass propagation of this recalcitrant forregeneration, economically important tree species.
