Abstract

The ptuτJOse of this study was to evaluate the cytotoxicity of lead [Pb(N03hl on NllI3T3 fibroblasts. Analysis of cytotoxicity was performed by MIT assay, NR assay and lactate dehydrogenase (LDH) activity after cells were trea때 with or without lead for 24 -72 hours. And also, light and electron microscopic studies were canied out to evaluate morphological changes of cells. The results were as follows : 1. Lead decreased the cell viability by MIT or NR assay on NllI3T3 fibroblasts in a dose• dependent manner after the cells were incuba뼈 with media of various concentrations of lead for 48 hours. 2. I0>J values of lead by the MIT assay and NR assay were 80 JlM and 60 JlM, respectively. These values stand for highly toxic on NllI3T3 fibroblasts according to the to잉 c criteria of Borenfreud et외‘ 3. The LDH activity increased in dose- and time dependent manner, and LDH잉 was determined at ~ 메[ Pb(NÜ3h 따ter cells were treated with 1 -100 JlM Pb(NÜ3)z for 48 hours. 4. In light microscopic observation, NllI3T3 fibroblasts were decreased in cell number, compared with the control 5. In electron microscopic observation, N퍼3T3 fibroblasts treated with Pb(N03)Z were shown cistemal dilatation of rough endoplasmic reticulum, many vacuole, some dense bodies in cytoplasm. From these res비ts， it is suggested that lead has cytotoxicity on NllI3T3 fibroblasts by showing as wel1 the decrease of cel1 viability, the increase of LDH activity, the decrease of cel1 number as cistemal dilatation of rough endoplasmic reticulum, vacuole and dense bodv in cytoplasm
