Abstract

Identification of urine sediments is not able to dip strip test. For determination of cell and crystal in urine we survey urine sediments by microscopy. In the most clinical laboratories take unstained wet smear microscopic test under bright field. But identification of cell and crystals is not enough under bright field. Therefor, we must do confirm test for identification of cell and crystal. In this report, we introduced several method that take effect in routine laboratory. The methods are as follows ; 1. By special microscope Phase contrast microscope is useful for identified cast and dysmorphic RBC with the changing of refractive index. Polarize microscope is useful for the oil droplets, oval fat body and crystals. For example, as property of cholesterol and cholesterol ester is birefringence , it showes maltese - cross pattern by using polarized microscope. Interferance contrast microscope is sterographic observation of the urine sediments. 2. By staining method Sternheimer stain(modified) method distinguish cells. Cytoplasm is stained red color with pyronin B, nuclear is stained blue color with alcian blue. Sudan m or Oil red - 0 dyes can stain oil droplets and oval fat body from orange to red color. Lugol stain method distinguish glycogen containing cells. Papanicolaou stain method is good for discrimination of cells and preservation of slides. 3. Determination of crystals in urine This method is utilization of specific character, solubility and observational typical shape.
