Abstract
Vitamin D is a hormone, not a vitamine. It can be ingested in the diet or produced in the skin as a result of exposure to sunlight. It is one of the most important bological regulators of calcium metabolism. Dietary and cutaneously formed vitamin D enters the circulation and is then bound to a vitamin D-binding protein. It is transported to the liver, Where it is hydroxylated to form 25-hydroxyvitamin D. 25-OH-D is formed in the liver, it is bound to the vitamin D-binding protein and is transported to the kidney. In the kidney, 25-OH -D is further hydroxylated to produce the bologically active metabolite, 1-alpha, 25-dihydroxyvitamin D. 1, 25-(OH)2-D promotes calcium absorption in the small intestine and increases the resorption of calcium from bone. Most of the earlier methods were generally cumbersome, frequently requiring several extractions and chromatographic purifications. In the past highly sophisticated techniques and, at times, instrumentation expertise were also required for successful implementation of vitamin D metabolite assays. However, recently reported direct methods using competitive protein-binding assay and radioimmunoassay capable for detection of vitamin D deficiency or excess. We’ve been used 3H -labelled kit in our lab, but it was not only inconvenient for usage but also got unsatified results because of its low precisions. In this study, we compared between 3H-labelled 125I-lavelled one. We evaluated the new3H-labelled 25-OH-vitamin D3 kit.
