Abstract

Heating of fonnalin-fixed paraffin-embedding tissue sections is often effective in restoring antigen reactivity lost during routine processing, but antigen retrieval(AR) procedures have become very popular. Immunohistochemistry procedures based on the high affmity of avidin and biotin, such as avidin-biotin-peroxidase complexes(ABC) procedure, are theoretically affected by endogenous biotin(EB), known to be present in some nonnal tissue, especially the liver, Kidney and adrenal cortex. Egg whites include Ovalburnin, Conalburnin, globulin, avidin and others. A vidin, exceptionally stable in egg whites, binds the biotin with a very high affmity. This study indicates that blocking of endogenous biotin reactivity by egg white treatment prevents any staining. Following microwave treatments, a simple streptavidin-peroxidase staining revealed retrieval endogenous biotin in normal tissues such as the liver, kidney and adrenal cortex, and carcinomas of the kidney and liver. Slides were the incubated with egg whites. Antigen retrieval(AR) can cause the reactivity of endogenous biotin(EB). Staining was prevented by the blocking of EB by egg white treatment. Retrieval of EB reactivity can cause pitfalls in diagnostic immunohistochemistry. but egg white treatment is an efficient method.
