성폭력 사건 증거물에서 DYS390 좌위의 특이 유전자형 분석 사례

2021년 2월 경기도 한 도시에서 술에 취한 피해자를 상대로 준강간 사건이 발생하였다. 성범죄 수사 절차에 따라 지역 해바라기 센터에서 피해자 신체를 대상으로 감정물을 채취하였다. 채취된 감정물은 성폭력 증거채취 응급키트에 포장되어 서울과학수사연구소로 긴급감정 의뢰되었다. 의뢰된 감정물에 대한 A-STR (상염색체 STR) 분석 결과, 피해자의 디엔에이형과 남성 디엔에이형이 혼합된 결과를 확보하였고, 혼합 디엔에이형에서 피의자 식별력을 증가시키기 위해 Y-STR (Y 염색체 STR) 디엔에이형을 추가 분석하였다. 그 결과 DYS390 좌위에서 null allele이 확인되었고, 인접 좌위인 DYS439에서 biallelic pattern이 검출되어 피의자의 DYS390 좌위가 allelic ladder를 벗어난 OMR (outside marker range) allele임을 추정하였다. DYS390의 OMR allele을 증명하기 위해 1차 분석에서 사용하지 않은 다른 Y-STR 키트를 이용한 분석 결과, DYS390에서 null이, 인접 좌위인 DYS438에서 동일한 경향의 결과를 얻었으며, 두 키트의 증폭 산물 크기를 allelic ladder와 비교하여 DYS390 좌위의 allele을 특정하였다. 본 사례를 통해 추후 발생되는 A-STR 및 Y-STR 분석 시 OMR allele 해석에 도움이 되고자 한다.
Abstract
In February 2021, a quasi-rape case occurred against a drunken victim in a city in Gyeonggi-do, South Korea. According to the sex crime investigation procedure, samples were collected from the victim's body at the local Sunflower Center, which provides integrated supports for victims of violence. Evidence was packaged in a sexual assault evidence collection kit and requested to the National Forensic Service Seoul Institute for urgent DNA analysis service. As results of Autosomal STR (A-STR) analysis, mixed DNA profiles of the victim and male were analyzed. In addition, Y Chromosomal STR (Y-STR) analysis was performed to increase suspect identification in mixed DNA. A null allele was observed at DYS390 and a biallelic pattern was observed at an adjacent locus, DYS439. It was estimated that the null allele was OMR (outside marker range) allele. In order to identify this, we analyzed using another Y-STR kit not used in the previous analysis. The same result was obtained at DYS390 and an adjacent locus, DYS438. The allele of DYS390 was determined through comparison to the product sizes of the two kits and the allelic ladder. We suggest that this case can be helpful for interpretation of OMR allele in A-STR and Y-STR analysis.
