Abstract
The purpose of this study was to examine immunocompetent cell numbers following eccentric and concentric muscle contraction in skeletal muscle. Eight-week-old Institute of Cancer Research (ICR) mice (n=84) were divided to eccentric muscle contraction group (ECC, n=42) and concentric muscle contraction group (CON, n=42). The tibialis anterior muscle was stimulated via the peroneal nerve to contract either eccentrically or concentrically (30 times × 5sets, Rest for 1 minute between each set). The tibialis anterior muscle was isolated before (pre, n=6) and 1 (n=6), 3 (n=6), 7 (n=6), 14 (n=6), and 28 d (n=6) after muscle contraction. Changes in the number of positive muscle fiber and immunocompetent cells (T, Th, Tc, neutrophils, and macrophages) were calculated by the histological analysis with HE stain and Immunohistochemistry. The number of positive muscle fiber was significantly higher (p<.05) in the ECC than in the CON at 3 and 7 d after muscle contraction. The number of Tc cells was significantly higher (p<.05) in the ECC than in the CON at 1, 3, and 7 d after the contractions. The number of neutrophils was significantly higher (p<.05) in the ECC than in the CON at immediately after, 1, 3, and 7 d after the contractions. The number of macrophages was significantly higher (p<.05) in the ECC than in the CON at all time course after muscle contraction. However, Changes in the number of T and Th cells were not significantly different between time course or groups. These results suggest that eccentric muscle contraction induces greater skeletal muscle damage and immunocompetent cells.
