Abstract
Purpose: This study investigated the effect of Smilax china ethanol extract (SCR) on improving anti-inflammatory and skin barrier function to confirm the potential of SCR as a natural material that can alleviate inflammatory skin diseases. Methods: The viability of macrophages (RAW 264.7 cells) and keratinocytes (HaCaT cells) following SCR treatment was assessed using the MTT assay. Anti-inflammatory activity was verified by confirming the inhibition of nitric oxide (NO) production, the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) proteins, and the expression of cytokine mRNA. Collagen synthesis was confirmed by enzyme immunoassay. The expression of matrix metalloproteinase-1 (MMP-1) protein was determined using Western blot. The effect of SCR on skin barrier function was assessed using reverse-transcription polymerase chain reaction (RT-PCR) assay targeting filaggrin, involucrin, and loricrin mRNA. Results: SCR significantly reduced NO production, the levels of proinflammatory cytokines (tumor necrosis factor-α, interleukin [IL]-1β, and IL-6), and iNOS and COX-2 expression in a concentration-dependent manner. SCR also inhibited collagenase and promoted type Ⅰ collagen synthesis in HaCaT cells. Furthermore, SCR decreased MMP-1 production and enhanced the mRNA expression levels of filaggrin, involucrin, and loricrin, which are essential for skin barrier function. Conclusion: Our findings suggest that SCR contributes to the symptomatic relief of inflammatory skin diseases by enhancing skin barrier function and exerting an anti-inflammatory effect. Therefore, SCR may be a potential functional material that can be used by the cosmetic industry to help alleviate atopic dermatitis.
