Abstract
The genetic diversity and spatial structures of natural populations of Cymbidium kanran onJeju island were estimated using ISSR markers. The genetic diversity(species level: h = 0.303,S.I = 0.389) and genotypic diversity(species level: GN/N = 0.884, D = 0.996) of this plant werefound to be higher than in other rare species. AMOVA analysis, revealed that 23% of the totalgenetic variation was allocated between two sub-populations. Taking the high levels of geneticdiversity and causative factors for differentiation into consideration, the major causes wereassumed to be the influence of alien individuals of C. kanran continuously introduced fromoutside in the past and the role of the intermediate host. Spatial autocorrelation analysisrevealed significant spatial genetic structure within 9m and 6m distance in two sub-populationsrespectively. As a sampling strategy for ex-situ conservation of C. kanran, a minimum distanceof 9m between individuals is recommended.
