초록

Efficient NADH regeneration from its oxidized form (NAD+) is required to support enzyme-based organic synthesis and the pharmaceutical industry. Prominent industrial NADH regeneration methods have shown promise; however, require the continuous addition of substrate, requiring downstream separation and purification. In this study, the utilization of diaphorase which can directly reduce the NAD+to NADH, with strong and stable mediator is presented which is capable of producing bioactive 1,4-NADH, faradaic efficiencies ~83% at varying temperature (20 to 60 °C). The efficiency of this approach as well as its high operational stability (87% of the maximum activity after six days) make it promising strategy for NADH regeneration.
