초록
The comet assay is a widely used technique for detecting DNA damage at the single-cell level. However, conventional systems are limited in throughput and often suffer from thermal instability and structural deformation during electrophoresis, especially when processing large sample volumes. In this study, we present the development of a novel high-throughput comet assay system specifically designed to address these limitations. The newly engineered electrophoresis tank features a compact footprint (316 × 579 × 71 mm) and incorporates an internal cavity for an embedded aluminum cooling plate to enhance thermal regulation. Additionally, the tank was constructed with reinforced MC nylon materials, including an 8 mm-thick external shell and a 6 mm-thick base plate, to ensure durability under repeated thermal cycling. The system supports the simultaneous processing of up to 40 slides, significantly increasing sample capacity compared to existing commercial setups. Functional validation using liver tissue exposed to the genotoxic compound ethyl methanesulfonate (EMS) confirmed clear detection of DNA damage, while vehicle-treated controls showed no signs of strand breaks. These findings demonstrate that the newly developed system offers reliable performance, thermal stability, and scalability, making it a suitable platform for high-throughput genotoxicity screening and research applications.
