초록
This study aimed to identify a Flavobacterium strain isolated from Atlantic salmon (Salmo salar) in Korea at the species level to provide foundational data for aquaculture disease management. The bacterium was isolated by culturing on Tryptic Soy Agar (TSA). The bactreria formed yellowish mucoid colonies on TSA and exhibited spreading yellow colonies on Cytophaga Agar. In the 16S rDNA-based sequence analysis for taxonomic identification, the isolate showed 99.49% identity to Flavobacterium bizetiae and 99.18% to F. saccharophilum. However, protein profile-based MALDI- TOF analysis result indicated either F. piscis or F. hibernum. To obtain a more accurate species identification, BLASTn and phylogenetic analysis were performed using six housekeeping genes in- cluding gyrB1, gyrB2, recA, rpoB, atpA, and trpB. The isolate exhibited the highest identities of 97.65%, 97.63%, 98.16%, 99.29%, 99.37%, and 95.85% to F. bizetiae, respectively, indicating the closest phylogenetic relation to this species, although it did not completely match the type strain. The low identities in gyrB1, gyrB2, and trpB may suggest those genes as high-resolution markers that reflect interspecific variation. These findings provide useful data for further study of species identification for Flavobacterium.
