Protoplast Culture by Electrofusion of Protoplasts from Solanum sisymbriifolium and Other Solanum Species

Abstract

betweenSolanumsis-ymbriifoliumandotherSolanumspecies(S.integrifoliumandS. toxicariumin S. sisymbriifolium,S. integrifoliumand the fused protoplast mixture; for theS. sisymbriifolium, a colony was observed in F me-dium(Kao mediumcontaining 5.0 mgL-1NAA, 1.0 mgL-12,4-D and 1.0 mgL-1BA); and for theS. integrifolium,inGmedium(ahalf strength MS mediumcontaing 0.03 Msucrose, 0.4 Mmanitol, 1.0 mgL-1NAA, 1.0 mgL-1kinetin) respectively. In mixedthe shoot and root formation rate, there was no diference between the parent of each breed and mixed protoplast regardless of themedium. Inthefusedprotoplast mixtureofS. sisymbriifoliumandS. toxicarium, acolonyformationwasalsoobservedinbothme-diaFandH(ahalf strengthMSmediumcontaining0.03Msucrose, 0.4Mmannitol, 0.1mgL-1NAA, 0.1mgL-1kinetin); andtherewas no difference in the shoot and root formation rate between the parent and the mixed protoplast.Key words -Solanumspecies, Culture media, Isozyme pattern
