Proteolytic enzymes have always been an essential necessity across various industries. Based on the growth characteristics of Mucoromycota, proteolytic enzymes were identified in its secretions. Among twenty previously unstudied species of Mucoromycota, Absidia spinosa was found to secrete proteases with azocasein hydrolyzing activity. Additionally, it was found to hydrolyze two specific substrates of proteases of human hemostasis system (S-2251, H-D-Val-Leu-Lys-pNA and S-2444, pGlu-Gly-Arg-pNA). To further investigate its growth characteristics and maximize the protease activity in its secretions, we conducted dynamic curve measurements using two fermentation media with different nitrogen sources. The results showed that the proteolytic ability of A. spinosa grown in fermentation medium 2 (containing amine nitrogen sources) was significantly higher than that in fermentation medium 1 (containing both amine and mineral nitrogen sources). Additionally, the growth morphology of A. spinosa varied slightly between fermentation media 1 and 2 with different nitrogen sources. Finally, we determined the optimal growth temperature and initial pH for A. spinosa.
