Abstract
The effects of chronic treatment with risperidone and haloperidol on the binding capacities of dopamine(DA) D2-like receptors were investigated in rat brain, the interesting regions were striatum and olfactory tubercle. To confirm the dose-dependent effects of usual and low doses haloperidol and risperidone, six groups of rats were treated with haloperidol or risperidone(control, 0.025, 0.05, 0.1, 0.25 0.5mg/kg/day N=6, each group) for four weeks. The effect of risperidone was compared with that of haloperidol in order to verify the region- specific changes of central dopaminergic system. Saturation analyses of the binding of [3H]spiperone to striatal and olfactorial DA receptor membranes were as follows, 1. There was significant proliferation in striatal membranes of haloperidol treated group(0.1, 0.25, 0.5mg/kg/ day), but no significant proliferation in olfactorial membranes. 2. There was significant proliferation in olfactorial membranes of risperidone treated group(0.025mg/kg), but no significant proliferation in striatal membranes. 3. The changes of dissociation constant(Kd) were not significant irrespective of brain regions and kinds of antipsychotics. These results indicate that the chronic treatment with haloperidol and risperidone induced the different change in central dopaminergic receptor. The atypical neuroleptics, risperidone, seemed to affect DA D2-like receptors differently in contrast to convential neuroleptics, haloperidol. This finding might suggest that the limbic system is the predilectional site of risperidone. The present findings might support the low dose strategy in the clinical treatment of schizophrenia and the regional selectivity of risperidone among the various central dopaminergic systems.
