Paternal Inheritance of Plastid Genome in the Interspecific Cross between Actinidia arguta and A. deliciosa

Abstract

Paternal inheritance of plastid genome was confirmed by PCR-sequencing in the interspecific cross between Actinidiaarguta and A. deliciosa. Both coding region and spacer region in the plastid DNA were PCR amplified, restriction digested, orsequenced. PCR-RFLP did not lead to diferentiation of the two parental species. However, it was possible to detect sequence dif-plastid inheritance was examined. The results showed that the plastid DNA of the progeny was identical to that of their paternalparent, A. deliciosa. Thus, the employed PCR-sequencing strategy proved an efficient and easy method to detect the presence ofspecific plastid sequence in the hybrids.
