Aim : To investigate whether leukocyte infiltration during neurogenic inflammation in the pulp is regulated by neuropeptides via inducing the release of proinflammatory chemokines interleukin-8 (IL-8) from human dental pulp , Methods : Primary human dental pulp tissue explants were stimulated with substance P (SP) , IL-8 produced in pulp explants was examined by immuno- histochemistry. Results : After the pulp tissues explants were cultured for 12 h and 24 h in the medium without FBS, IL-8 were expressed only around the cell rich zone and blood vessels in the pulp tissues , SP(10-4 M) induced lL-8 expression in the pulp tissue explants after 24 h of SP stimulation, while TNF-a(20ng/ml) did not induce the IL-8 in the pulp tissue. The stronger lL-8 staining was detected in the pulp tissue explants after 6 and 12 h of TNF-a(20ng/mI) stimulation, while only dead cells were revealed in the pulp tissue after 24 h of TNF-a(20ng/mI) stimulation. Conc1usions : SP significantly induces IL-8 expression in human dental pulp suggesting an important role of SP in recruiting neutrophils in localized pulp tissue , SP may have positive relation with the inflammation of the human dental pulp tissues.
