Abstract
In recent, CBC count is measured by the autoanalyzer of the blood cell count in many routine laboratory, but some cases that is out of reference value showing incorrectly result. This study was carried out to elucidate differences between morphological findings and blood cells autoanalyzer in the specimens that appeared out of reference value, using light microscopy with blood smear treated Wright’ s stain and scanning electron mlcroscopy. We observed that the results of the Cell-Dyn 3000®, direct blood smear, and SYSMEXSE-9000 . are not same as those of each in the processing samples. The present study shows that RBC is composed of target RBC on the capillary bloodsmear test. The main reason that pseudoleukocytosis appeared to be result of the equipment of Cell-Dyn 3000® is caused by falsely increased numbers of lymphocytes. RBC was not identified at both light microscopy and scanning electron micrescopy, after sample which is easily lysed by the solution of the sheath was immediately admixed with the solution of the sheath. However, after "resistant RBC" in the specimen that showed pseudoleukocytosis is admixed with the solution of the sheath for 30 seconds, RBC was still observed at to light microscopy and scanning electron microscopy.
