초록
We determined the allele frequencies and forensic parameters for 21 STR autosomal loci in order to improve discriminatory power in the national database. Currently, DNA profiling for individuals in the South Korean population is performed using the AmpF/STR Identifiler PCR Amplification kit (Applied Biosystems) including the CODIS core STR loci. In order to prepare for the new requirements for the extension of STR loci, we have undertaken to genotype DNA samples from a population sample of 250 unrelated individuals living in South Korea. The PowerPlex ESI 17 System (Promega) configuration contains 8 loci as CODIS (D3S1358, D18S51, D16S539, D21S11, vWA, TH01, FGA, D8S1179), 6 new recommended loci (D22S1045, D2S441, D10S1248, D1S1656, D12S391, SE33) and the amelogenin sex marker. The most discriminative locus was SE33 (0.988) while the least was TPOX (0.765). The combined probability of identity (PI) was 9.66 × 10−25 and the combined mean exclusion chance was 0.999999993. For 21 autosomal STR loci, no deviation from the Hardy-Weinberg equilibrium was observed with the exception of the CSF1PO locus (P > 0.05). After employing the Bonferroni correction for the number of loci analyzed, this single p value gives no reason to reject the null hypothesis. The concordance study made for this population data only revealed one discordant allele (TH01) between the kits. Full concordance between the typing results for the two kits was observed in 99.99% of the STR allele calls compared. These results indicate that these 21 loci are useful genetic markers for forensic personal identification, and for paternity testing in the Korean population.
