Abstract
The application of multiplex STR kit results in a full DNA profile in the majority of high-quality DNA samples. However, the increased demand to analyze limited DNA samples from a low copy number or skeletal remains increases the number of samples where only a partial DNA profile is obtained due to lowquantity DNA and/or degradation of the DNA. In an effort to increase the amount of information derived from these types of DNA samples, the AmpFlSTR^® Minifiler^(TM) PCR Amplification kit has been developed. Internal validation of the Minifiler^(TM) kit revealed no differences for the STR loci while the sensitivity study showed a reproducible DNA profile with as low as 0.063 ng of Human DNA control 9947A. The full profile of a minor peak could be detected in a 1:3 of the mixture. This article describes the internal validation performed according to the SWAGDAM revised guidelines. Based on the results obtained, guidelines for the interpretation of DNA analysis procedure and DNA STR profiles drawn up to use the Minifiler^(TM) kit in forensic casework at this laboratory.
