Abstract
As the worldwide population of companion animals increases, dog-related crimes are also increasing, and it has reminded the importance of dog-related biological evidence. Also, these cases tend to be a frequent mix of human and dog samples, making precise identification more difficult. In this study, we developed a method using droplet digital PCR (ddPCR) to simultaneously identify and quantitatively detect human and dog DNA in mixed samples. HLA-DRA and OR6D7 were chosen as specific markers for human and dog respectively. The species specificity of each target was confirmed using 14 control DNA samples including 11 mammals and 3 birds. Sensitivity testing revealed detection limits of 0.0098 ng/μL for human and 0.0195 ng/μL for dog targets. Moreover, Even in samples containing two types of DNA mixed, each DNA was accurately detected independently. Additionally, the presence of certain inhibitors did not impact the analysis. Consequently, this study presents a reliable method for the simultaneous and precise identification and quantification of human and dog DNA, offering promising possibilities for advancements in forensic DNA analysis and related fields.
