초록
In mutation breeding, ionizing radiations are widely used as physical mutagens. Protonbeams (PBs) are known to have physically intermediate properties between low linear energytransfer (LET) gamma rays (GRs) and high LET ion beams, but their mutational effects havestill not been characterized. This study was conducted to estimate the biological effect of protonbeam irradiation compared with gamma irradiation and investigate the optimal dose for mutationinduction in rice. Rice seeds were irradiated using a 100 MeV Linear Accelerator (TR103) at theKorea Multi-Purpose Accelerator Complex for PBs and a 60Co gamma irradiator at the AdvancedRadiation Technology Institute for GRs, with doses of 50, 100, 150, 200, 250, 300, 400, 500, 600,and 700 Gy in both cases. The PB irradiated seeds did not germinate from 500 Gy or higherdoses, whereas the GR irradiated seeds did not germinate from 600 Gy or higher doses. The doseshowing a half germination rate against the control was 200 Gy and 350 Gy for PBs and GRs,respectively. The median lethal dose and the median reduction dose (RD50) values estimated byanalyzing the survival rates, and growth of the four-week-old seedlings was 150 Gy and 175 Gyfor PBs, and 250 Gy and 300 Gy for GRs, respectively. These results suggest that the biologicaldamage of PBs is more severe than that of GRs at the same dose. To induce mutations using PBsin rice, doses from 100 to 150 Gy seem to be proper.
