Abstract
The purpose of this study was to compare the characteristics and irritation potentials of three commercially available 3D skin models (EpiDermTM SIT, LabCyte EPI-24 and KeraSkinTM-VM) that use human keratinocytes as an alternative means of testing skin irritation. The test substances were applied on the surface of insert cells after pre-incubation (24 h). The test substances were completely washed, and then models were incubated for 48 h. Cell viability was measured using the MTT assay to measure levels of skin irritation. Quality control indices, such as barrier function (TEER), absorbance of negative control, cell viability of positive control, absorbance of extraction buffer used in MTT assay, acceptability range (upper and lower limit), and results range and consistency between inserts, were in agreement with the acceptance criteria. With the exception of 4-methylthio-benzaldehyde (negative substance in globally harmonized system (GHS)), which led to false positives in all 3 models of the preliminary study, other test substances showed consistent results with the GHS for classification standards in OECD Guideline 439. Taken together, results for sensitivity, specificity, and accuracy of the three models were 100, 67, and 83%, respectively; these values are on par with OECD Guidelines.
