Measurement of the expression of Wnt3a, Wnt5a, interleukin (IL)-1α, and transforming growth factor (TGF)-β in human dermal papilla cells (hDPCs) was performed for evaluation of the hair growth promotion and hair loss inhibitory efficacies of Stephanandra incisa ethanolic extract (SIEE). Treatment with 25 μg/mL SIEE and 1 μM minoxidil (MXD) resulted in upregulated expression of Wnt3a mRNA by 6.0% and 62.7%, respectively, and expression of Wnt5a mRNA by 46.3% and 70.3%, respectively. Treatment with 25 μg/mL SIEE and 1 μM MXD resulted in downregulated expression of IL-1α mRNA by 31.7% and 42.7%, respectively, and expression of TGF-β mRNA by 31.3% and 46.3%, respectively. SIEE was less effective than MXD; however, treatment with SIEE resulted in upregulated gene expression of hair growth factors, Wnt3a and Wnt5a, and downregulated gene expression of hair loss factors, IL-1α and TGF-β. These results suggest the potential for use of SIEE as a natural cosmetic material for promotion of hair growth and inhibition of hair loss. Measurement of the antioxidative efficacy was performed in a cell-free system. The XO inhibitory activity of S. incisa methanolic extract (SIME) and SIEE at 1,000 μg/mL was 61.3% and 57.1%, with an IC50 of 407.7 and 476.5 μg/mL, respectively. The SOD-like activity of SIME and SIEE at 1,000 μg/mL was 59.8% and 73.8%, with an EC50 of 585.5 and 431.6 μg/mL, respectively. The electron-donating ability of SIME and SIEE at 1,000 μg/mL was 80.1% and 83.7%, with an SC50 of 96.0 and 49.4 μg/mL, respectively. SIEE exhibited higher antioxidative efficacy than SIME. High efficacy for reduction of reactive oxygen species may be an underlying mechanism of downregulation of the gene expression of hair loss factors by SIEE in hDPCs.
