Abstract
With the increase in environmental pollutants and oxidative stress, there is an increased interest in identifying natural bioactive compounds with antioxidant and skin-protective properties. Our study investigates the protective effects of the ethanol extract of the edible insect Gryllus bimaculatus-Cordyceps militaris against oxidative stress- induced damage on human keratinocytes (HaCaT). The extract was prepared using 70% ethanol and the phenolic and flavonoid contents were analyzed. Antioxidant activity was evaluated through (2,2-diphenyl-1-picrylhydrazyl) DPPH and 2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) ABTS radical scavenging assays. Cytotoxicity was assessed using an (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) MTT assay on HaCaT keratinocyte cells. Potential anti-aging properties were determined by measuring the inhibitory effects on elastase and collagenase through the analysis of the expression of matrix metalloproteinase-1 (MMP-1) and hyaluronic acid (HA) synthesis. The extract contained significant amounts of total phenolics (26.62 mg GAE/g) and flavonoids (0.372 mg CE/g). It exhibited strong radical scavenging activity (DPPH: 62.75%, ABTS: 40.51%). There was no cytotoxicity at tested concentrations in the extract with effective inhibition of elastase (42.6%) and collagenase (52.1%) activity. MMP-1 expression was significantly reduced and HA synthesis was enhanced by up to 90%, which suggested improved skin hydration. Our findings establish that the Gryllus bimaculatus-Cordyceps militaris extract possesses antioxidant, anti- aging, and moisturizing effects, which makes it a promising candidate for natural skincare applications.
