Abstract
In the present study, the effect of treadmill exercise on chronic intermittent cold stress-induced apoptotic neuronal cell death in the hippocampus of mice was investigated. Male ICR mice were divided into four groups: the control-rest group; the control-exercise group; the cold-rest group; and the cold-exercise group (n = 5 in each group). In the present study, the chronic intermittent cold stress was induced by exposure of mice to cold room at 4℃ for 3 h once a day, 6 times per week during 3 weeks. The treadmill exercise load consisted of running at a speed of 5 m/min for 30 min once a day, 6 times per week for 3 weeks. Immunohistochemistry for caspase-3 and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL) staining were performed to determine apoptotic neuronal cell death in the hippocampal dentate gyrus. Our results revealed that the expression of caspase-3 and the number of TUNEL-positive cells were significantly increased by cold stress (p<.05), and treadmill exercise significantly suppressed the expression of caspase-3 and the number of TUNEL-positive cells after cold stress (p<.05). The findings of our study provide evidence that treadmill exercise may ameliorate the brain damage induced by cold stress.
