Abstract

Cell adhesion is well known to have various effects on cellular activities including apoptosis, cell cycle control, migration, and differentiation. In the present study, the effect of substrate on osteoclast differentiation was studied, First osteoclast differentiation from bone-marrow derived mononuclear cells under the influence of RANKL and M-CSF was examined on the apatite thin film or collagen matrix. Osteoclast differentiated well on the collagen matrix and apatite thin film of fine surface roughness. However, no osteoclast was differentiated on the rough surface of apatite thin film. Next the effect of substrate conditions on the mRNA expression of molecules such as RANKL, M-CSF, OCIF, 江1, TNF-a IFN-Y, TGF냉, and osteopontin which are relevant to osteoclast differentation, was examined in fibroblast or os않oblast. RANKL and M-CSF were expressed in primary cells of osteoblast and fibroblast. However, IFN-r and TGF -ß were not expressed in those cells. Furthermore there was a difference in expression of mRNA de야nding on the substrate conditions. These results indicate that substrate where precursor cells of osteoclast attach can control the differentiation of osteoclasts, In addition, osteoblast or fibroblast can modulate differentiation of osteoclast depending on the attaching conditions.
