Abstract
Thrombospondin-l (TSP-l) is a major component of platelet Q-granules and megakaryocytes. TSP- l has bæn shown to bind to matrix proteins, plasma proteins, cytokines, growth factors, and proteases. Several results suggest that TSP-l is important mediators of bone metabolism. The present study was undertaken to deterrnine the possible cellular mechanism of action of TSP-l in bone metabolism‘ The effects on the osteoblasts were determined by meaSl띠ng 1) cell viability, 2) alkaline phosphatase (ALP) activity, 3) gelatinase activity, and 4) NO production. The effects on the osteoclasts were investigated by measuring 1) tartrate-resistant acid phosphatase (+) multinucleated cells formation, and 2) resorption areas after cultwing osteoclast precursors TSP-l treatment showed a significant increase in both the viability and ALP activity of osteoblasts. TSP-l increased the ac디vity of gelatinase. Treatment of TSP-l had no effect on the production of NO from osteoblasts. 1n addition, TSP-l had no effect on the osteoclast generation induced by macrophage-colony stimulating factor (M-CSF) and receptor activator of NF-kB ligand, but enhanced osteoclastic activity in M-CSF-dependent bone manrow macrophage cell cultures. Taken these results, TSP-l may be a reg비atory protein within the bone marrow rnicroenvironment.
