Abstract
To investigate the function of the active components in chili peppers (Capsicum annuum L.), which are used as riot-control agents, capsaicin analogs (1-8) were used as substrate molecules for molecular docking against 2 receptors (CYP2E1; PDB 3GPH and CYP2C9; PDB 1R9O) in the human body. In addition, the roles of the 2 receptors (CYP2E1 and CYP2C9) of pepper capsaicin analogs (1-8) were investigated in the correlation between molecular docking score (DS) and Scoville heat units (SHU), a measurement indicating the spiciness of the chili pepper. Based on the volume (CYP2E1; 112 Å3 and CYP2C9; 118 Å3) of the reaction site, CYP2C9 showed a larger volume of about 6 Å3 more than CYP2E1. Among the capsaicin analogs, homocapsaicin (5) showed the largest docking score, and CYP2C9 tended to have a higher docking score than CYP2E1 (DS:CYP2E1; 44.54 and CYP2C9; 55.37; ΔDS=10.83). In conclusion, the structure-activity relationship (SAR) between DS and SHU revealed that the capsaicin analogs (1-8) selectively undergo dehydrogenation (oxidation and reduction) with CYP2E1.
