Abstract

This study was conducted using response surface analysis to confirm the optimal extraction conditions for efficient supply of verbascoside and rutin, which are indicator components of Abeliophllum distichum Nakai., which is being developed as a resource plant. An experiment was performed on 17 sections using hot air dried leaves of A. distichum by setting three independent variables (ethanol concentration, extraction time, extraction temperature) and the contents of verbascoside and rutin were measured. The optimal extraction conditions for verbascoside were 60% ethanol concentration, 14.4 h extraction time, and 75°C extraction temperature. The expected value of verbascoside under optimal extraction conditions was 193.92 ㎎/g, and the experimental value was 193.66±4.37 ㎎/g. The optimal extraction conditions for rutin were 52% ethanol concentration, 30 h extraction time, and 75°C extraction temperature. The expected rutin content under optimal extraction conditions was 13.67 ㎎/g, and the value obtained through the actual experiment was 14.38±0.31 ㎎/g. It was confirmed that verbascoside and rutin could be efficiently secured through optimal extraction conditions derived by response surface analysis.
